Reveal® Q+ for Aflatoxin EndPoint is a lateral flow immunochromatographic assay based on a competitive immunoassay format intended for the quantitative testing of aflatoxin in corn using the Raptor® Solo Platform.

Aflatoxin is a toxic and carcinogenic substance produced by certain strains of the molds Aspergillus flavus and A. parasiticus. There are four principle types of aflatoxin: B1, B2, G1 and G2. Aflatoxin B1 is the most frequently encountered of the group and the most toxic. The commodities most affected by aflatoxin are corn, peanuts, cottonseed, milo, and the majority of tree nuts.

The effects on animals that ingest excessive amounts of the toxin range from chronic health and performance problems to death. Aflatoxin has been shown to cause liver damage or cancer, decreased milk and egg production, immune suppression, and interference with reproductive efficiency.

Many countries have set maximum allowable levels of aflatoxin in food and feed. Therefore, accurate determination of the toxin’s presence is crucially important to those monitoring the quality of food and feed in which aflatoxin may occur. Testing these commodities for the toxin requires careful sampling, chemical extraction, sanitation, and quantitative analysis.

Technologie

In the test, the extract is wicked through the reagent zone, which contains antibodies specific for aflatoxin conjugated to colloidal gold particles (gold complex). If aflatoxin is present, it will be captured by the gold complex. The gold complex, along with any free gold-complex, is then wicked onto a membrane, which contains a zone of aflatoxin conjugated to a protein carrier. This zone captures any unbound gold complex, allowing the particles to concentrate and form a visible line. As the level of aflatoxin in a sample increases, free aflatoxin will bind with the gold complex allowing the less gold complex to be captured in the test zones. Therefore, as the concentration of aflatoxin in the sample increases, the test line density decreases.

Algorithms programmed into the readers convert the line intensities into a quantitative result displayed in parts per billion (ppb). The membrane also contains a control zone where an immune complex present in the reagent zone is captured by an antibody, forming a visible line. The control line will always form regardless of the presence of aflatoxin, ensuring the strip is functioning properly.
