Pseudomonas Agar Base is used for the selective and differential isolation of Pseudomonas sp. using CN or CFC supplementation in a laboratory setting. Pseudomonas Agar Base is not intended for use in the diagnosis of disease or other conditions in humans. Pseudomonas Agar Base was developed to be supplemented with CN (Cetrimide & Sodium Nalidixate) or CFC (Cetrimide, Fucidin, & Cephaloridine). This base formula is a modification of King's A Medium. The CN Supplement is recommended for the isolation of Pseudomonas aeruginosa, where the addition of Sodium Nalidixate and the reduction of Cetrimide improved recovery. Pseudomonas Agar Base with the addition of CN Supplement, demonstrated enhanced pigment formation of P. aeruginosa and increased inhibition of Klebsiella, Proteus, and Providencia spp. Pseudomonas Agar Base with the addition of CFC Supplement is recommended for the selective isolation of Pseudomonas spp. Mead and Adams reduced Cetrimide, permitting growth of all pigmented and non-pigmented psychrophilic pseudomonads. The antimicrobics, Fucidin and Cephaloridine, were added to increase the selectivity of the medium. This formulation is used for Pseudomonas spp. from chilled foods and processing plants. Burkholderia cepacia (formerly Ps. cepacia) is isolated from water sources, and can be problematic in the preparation of sterile products for the pharmaceutical, food, and cosmetic industries. Pseudomonas Agar Base is recommended by ISO for the enumeration of Pseudomonas spp. from meat and meat products.

Preparation
  1. Dissolve 48.4 g of the medium and 10 mL of Glycerol in one liter of purified water and bring to a boil.
  2. Autoclave at 121°C for 15 minutes.
  3. Cool to 45 – 50C before adding supplements.
