Technology

Assay Principle

Reveal for Histamine is a single-step lateral flow assay based on a competitive immunoassay format. The extract is wicked through a reagent zone, which contains histamine-specific antibodies conjugated to colored particles. If histamine is present, it will be captured by the particle-antibody complex. The histamine-antibody-particle complex then is wicked onto a membrane, which contains a zone of histamine conjugated to a protein carrier. This zone captures only uncomplexed antibody-particle conjugate, allowing the colored particles to concentrate and form a visible line. The membrane also contains a control zone where the antibody-particle complex present in the reagent zone is captured by an antibody, forming a visible line. The control line will always form regardless of the presence of histamine, ensuring the strip is functioning properly. In addition, the control line also is used as a reference for comparison to the test zone for visual interpretation of the test results. If histamine is present above a pre-determined limit, a line lighter than the control line or no line will form. Therefore, a positive sample will result in a light line or absence of a line in the test zone. Alternatively, a negative sample will form a line of the same or darker intensity as the control line in the test zone.
